أهلا وسهلا بك عمر وبالمستمعين الأعزاء، من سعادتي ان اتواجد في هذا البودكاست وأن اتواصل مع متابعي العلوم الحقيقية.
شكراً جزيلاً لحضورك، وانا متشوق للبدء بالموضوع، الفكرة الأولى او السؤال الأول لدي هو ما هي قصة الميتوكوندريا، نرى أن هناك حمض نووي للميتوكوندريا، يبدو الأمر وكأن هناك دولة داخل دولة، خلية داخل خلية؟ فما هي القصة؟
جميعنا نذكر في كتب العلوم ما يسمى ببيوت الطاقة أو المتقدرات او الميتوكوندريا بحسب اختلاف التسميات وهي واقعا جزء من العضيات الموجودة في الخلايا. يعود اكتشافها الى سنة 1857 على يد العالم البيرت فون كوليكر (Albert von Kölliker). ظهر تحت المجهر في حينها ان هناك شيء اشبه بالخيوط. لكن هذه الخيوط بقيت مجهولة حتى سنة 1898 عندما أعطيت الاسم ميتوكوندريا على يد العالم كارل بندة (Carl Benda). وتسمية ميتوكوندريا تعني المتقدرات الخيطية. والميتوكوندريا كما يوحي اسمها بالعربية هي المركز الذي تصنع به اهم جزيئة او العملة للطاقة في الجسم وهي الادينوسين ثلاثي الفوسفات (ATP). والاي تي بي هو بمثابة العملة في الطاقة للعمليات الحيوية المختلفة، مثلما ان الدولار في العالم اليوم هو العملة في الكثير من العمليات الاقتصادية.
أصل الميتوكوندريا بقي مجهولاً حتى جاء آلتمان (Richard Altmann) الذي بحث في أصول الميتوكوندريا، وبعد تحليله لشكلها - في وقت لم تتوفر فيه الأجهزة الحديثة اللازمة – اسماها بالخيوط الشبيهة بالأحياء المجهرية (bioblasts) أو الأرومات الحيوية. وأصل الميتوكوندريا حتى الآن يقترب من ملاحظة آلتمان في ذلك الوقت وهي كونها شبيهة بالأحياء المجهرية أو كأنها خلية داخل الخلية، فهي قد تكون خلية هاجمت خلايا الكائنات الحية متعددة العضيات في وقت ما في تاريخ سحيق. وفي العام 1963 حاول كل من مارغريت وسيليفان ناس (Margit and Sylvan Nass) ان يفهما الميتوكوندريا في أجنة الدجاج، وقد رأوا أن هذه العضيات تحمل حمضاً نووياً خاصاً بها، وهنا كان الاكتشاف الكبير بأن لهذه العضيات حمضها النووي الخاص بها المنفصل عما موجود في النواة الاعتيادية.
هناك مجال آخر قد يعلمون بهذه القصة، وهم المختصون بفحوصات الحمض النووي الذين يستخدمون هذا الحمض النووي في تتبع سلالة الأمهات، فهل هو قادم من الأم؟
المميز بهذا الحمض النووي هو أننا نرثه عن طريق الأم فقط. وهناك نظريتان لتفسير ذلك، الأولى ما يصب حول أن استنساخ الحمض النووي للميتوكوندريا يكون محدوداً جداً اثناء تكوين النطف، اما الثانية فهي أنه رغم قلة الميتوكوندريا في خلايا الحيوانات المنوية فإن هناك آلية خاصة لتكسير الحمض النووي القادم من ميتوكوندريا الرجل بعد التخصيب. وبذلك يمكن للعلماء أن يتتبعوا خط وراثة الأمهات عبر الحمض النووي للميتوكوندريا ومعرفة أنها قادمة من الأمهات وهناك تسمية شعبية للميتوكوندريا (عضية حواء) لأنها آتية عن طريق الأم فقط. رغم ذلك، حتى الان الدراسات لم تظهر السبب الرئيسي لوجود الميتوكوندريا من الأمهات فقط. لكن لو حللنا مقدار بيوت الطاقة بين النطف والبيوضات، لوجدنا بحدود 100 ألف نسخة من الحمض النووي للميتوكوندريا في البويضات بينما في النطف هناك فقط خمسة آلاف نسخة. إذاً هناك نوع من السيطرة من خلال العدد. تفسير وجود هذا العدد الكبير من نسخ الحمض النووي هو عدم وجود آلية لإصلاح الحمض النووي في الميتوكوندريا فنجد ذلك التراكم في تلك النسخ. لذا وللسيطرة على الطفرات الوراثية في الحمض النووي للميتوكوندريا صار هناك نوع من الانتقال الأحادي للجينات والذي يمكن تفسيره أيضاً بنشوء الميتوكوندريا التي نجدها شبيهة بخلايا البكتيريا البدائية وهذا ما يعزز الفرضية بأنها كانت خلية اقتحمت الخلية الحية، وصارت جزءاً منها.
