王道還科普專欄〈意外:機會之窗〉全文朗讀 高三畢業那年5月,也就是聯考前兩個月,我買了一本書,書名《科學探奇》,是翻譯書。書裡有十個科學史故事,作者都是說故事的高手,我讀得津津有味,悠然神往,忘了聯考。印象最深刻的一篇,吸引我的不是內容,而是它的第一個句子:科學研究上的重大興奮事情之一是:你出去獵兔子,你偶爾會捉到一隻熊。這句中文有點彆扭,是翻譯體,但是意思很清楚:有些重要的科學發現,是意外的驚喜。書裡的故事大部分淡忘了,包括這一篇,但是這一句我一直記得。科學史上,的確有不少意外的重大發現。有人蒐集成書,讀了不但益智、怡情,也是教學、聊天的好材料,最能吸引自命不懂科學的人注意。那些事件若以「無心插柳」形容,未免陳腔濫調,「出門獵兔逮到熊」就神了。也難怪,作者是大名鼎鼎的艾西莫夫(Isaac Asimov, 1920-1992)。可是同樣的故事多講了幾遍,驚喜不免退燒,便可能注意到有些意外根本不是意外,而是疏忽。實驗室裡最容不得的就是疏忽。要是疏忽居然成為美談,我們怎麼教學生、要求學生呢?英國生物冷凍技術開拓者派克斯(Alan S. Parkes, 1900-1990)的重大發現就是一個例子。其實派克斯的成就,就他中學的學業表現而言也是個異數。他在第一次世界大戰快結束的時候入伍,不久德國便投降。這一當兵資歷使他有幸免試進入劍橋大學,成為達爾文的學弟——90年前,達爾文也進了劍橋的基督學院。派克斯畢業後到曼徹斯特大學念博士,開始建立自己的學術生涯。他生逢其時,參與了生殖生理學的發展,成為領導人物。 1951年,以解凍精子授精的卵成功孵化為小雞二次世界大戰後,派克斯改行研究冷凍生物學(cryobiology),研發長期保存動物精子的技術。這種技術對於人類生殖與畜牧業都提供了全新的願景。1948年秋,派克斯的團隊開始實驗以果糖保護冷凍家雞精子的技術。由於結果並不令人鼓舞,就暫時放手,將許多調製好的溶液全都收進冷藏櫃,打的主意不外是:事緩則圓。幾個月後,研究人員恢復實驗,一瓶瓶使用那些溶液,結果其中有一瓶居然能使精子在攝氏零下79度保有幾乎十足的游動能力。起先研究人員以為那瓶溶液起了化學變化,因為瓶裡已長滿了黴,於是惴惴不安地吸取了10幾c.c.做化學分析。結果瓶裡根本不是果糖溶液。原來那是製作玻片標本用的固定液,成分是甘油、蛋白。進一步分析後,確定甘油是有效成分。那是組織冷凍技術的重大突破,至今仍然使用。然後研究人員發現:精子解凍後要是將甘油清乾淨,就能恢復雄風——使卵子受精。1951年,以解凍精子授精的卵成功孵化為小雞,為生殖生物學樹立了一座里程碑。這個故事乍聽之下有傳奇的魅力,難題、難關、幸運俱全。但是仔細一琢磨,就會發現不可原諒的疏忽:研究人員居然沒有將自己配製的試劑貼上標籤!哪個實驗室會容許這樣的疏忽?另一個現象更不可思議:溶液瓶裡長了黴!實驗的關鍵試劑怎麼可以受到污染?「出門獵兔逮到熊」,英文裡有一個字,專門指這種意外:serendipity。這個字的字源是地名,指印度洋裡的錫蘭,現在叫做斯里蘭卡。對歐洲人來說,那個「遠東」地名暗示著:意外產生美妙的結果,其中必然有神祕的智慧。那麼派克斯實驗室裡的疏忽/意外,透露了什麼智慧?也許另一個更有名的科學意外能協助答覆這個問題。 弗萊明注意到青黴周緣的細菌已經溶解,就不是意外了 1928年9月,英國細菌學者弗萊明度假歸來,回到倫敦的實驗室,注意到他打算清洗的一個葡萄球菌培養皿裡有一塊青黴(一種真菌),緊鄰青黴周緣的細菌菌落已經溶解。弗萊明懷疑那是青黴菌分泌的抗菌物質造成的,於是他將黴菌菌種保留下來,專門做實驗測試它的抗菌能力。結果弗萊明發現了青黴素——大家熟悉的名字是盤尼西林——因而獲得1945年諾貝爾生醫獎。後來學者才知道,弗萊明採集的青黴菌不但是罕見的物種,而且是罕見的變種,因此真菌專家都未必熟悉。那一青黴菌種飄入弗萊明的實驗室純屬意外。但是弗萊明注意到青黴周緣的細菌已經溶解,就不是意外了。原來第一次世界大戰之後弗萊明就以搜尋無毒的抗菌物質為研究重心。某天,他因為感冒而鼻水直流,突發奇想,將鼻水滴入細菌培養皿中,意外發現鼻水含有可以抑制細菌的「溶菌酶(lysozyme)」。1922年,弗萊明將溶菌酶的實驗結果寫成論文發表,提到鼻水、眼淚、雞蛋、水果等常見物品中都有溶菌酶。可惜溶菌酶只能抑制無害的細菌,奈何不了葡萄球菌、鏈球菌等常見害菌。此外,弗萊明的化學功力不足,他知道哪些物質中含有溶菌酶,卻無法將它純化,做進一步的定性、定量研究。總之,青黴在細菌培養皿裡造成的景象,弗萊明並不陌生,才能將意外轉化為驚喜。 有行動,就可能發生意外 巴斯德說,「幸運女神青睞有準備的人」。那一年(1928)獲得諾貝爾生醫獎的另一位法國細菌學者尼科勒(Charles Nicolle, 1866-1936)更積極,他說:幸運女神只青睞懂得如何追求她的人。不過,發現盤尼西林的故事並不止於法式浪漫。無人不知盤尼西林是意外的發現,但是真正的意外卻少有人注意,特別是喜愛科學意外故事的人。原來我們當作抗生素的化學物質,對於微生物而言,也許並不是抗菌物質。這樣理解抗生素,才能解釋兩個重要事實:一、在自然界,會製造抗生素的微生物很少;二、在自然界,會製造抗生素的微生物,產量太小,不足以創造生存空間;推論:在自然界,微生物不依賴我們稱之為抗生素的物質互相對抗。因此在自然界搜尋抗生素才越來越困難。重大的科學發現,來自疏忽也罷、意外也罷,暴露了我們自身的侷限。我們對大自然若沒有成見,就無法進行系統的觀察。可是人的理性有限,再高明的心智也不足以觀照全局,對於宇宙真相只能提供一二快照。因此意外才會在科學史上製造了那麼多佳話。意外提供了破壞式創新的機會。歸根結柢,一切源自求知的欲望、以及它激發的行動;有行動,就可能發生意外。你出去獵兔子,你偶爾會捉到一隻熊。 作者小傳─王道還台北市出生,從小喜歡閱讀,但是從未想過寫作,因為小學五年級投稿國語日報兩次皆遭退稿。大學三年級起意外接到翻譯稿約,以後寫作亦以翻譯為起點(意思是抄襲)。在思想上,對於「思考」產生全新的認識,是在高二暑假讀了《西洋哲學史話》(台北:協志工業出版)、《相對論入門》(香港:今日世界出版社)兩本書。從高一起就對演化生物學發生興趣,後來以生物人類學為專業可能並非偶然,可是對科學史、科學哲學的興趣從未間斷。 下載「鏡好聽」APP,聲音節目隨身聽~ 最多獨家更新內容,請下載《鏡好聽》APP:https://mirrormediafb.pros.is/LY67K
