Charakterisierung des HLA-Klasse-I Ligandoms in Primärtumoren beim humanen Nierenzellkarzinom
T-Zellen können tumorassoziierte HLA-präsentierte Peptide auf Tumorzellen des Nierenzellkarzinoms erkennen, und diese dann eliminieren. Gelingt es, diese Fähigkeit des Immunsystems mit einer Impfung geziehlt anzuregen kann eine Peptid-basierte Immuntherapie ein Verfahren zur Behandlung des Nierenkrebeses darstellen. Aus diesem Grund wurden durch massenspektrometrische Untersuchungen HLA-Klasse-I Ligandome maligner transformierter Zellen des soliden Nierenzellkarzinoms isoliert. Mit Hilfe von vergleichenden Genexpressionsanalysen wurden diejenigen Gene identifiziert, die aufgrund ihres Expressionsmusters als tumorassoziiert angesehen werden können. Diese aus HLA-Liganden abgeleiteten Tumorantigene stellen potentielle T-Zellepitope für eine Peptid-basierte Immuntherapie dar.

Immungenetische Untersuchungen zur Assoziation von Polymorphismen im nicht-klassischen MHC-Klasse-Ib-Gen HLA-G mit der schwangerschaftsinduzierten Autoimmundermatose Pemphigoid gestationis
Pemphigoid gestationis ist eine seltene blasenbildende Autoimmundermatose unklarer Ätiologie, die ausschließlich im Zusammenhang mit Schwangerschaften oder schwangerschaftsassoziierten Tumoren beobachtet wird. Als Autoantigen konnte das Transmembranprotein Kollagen-Typ-XVII, ein Strukturelement der Hemidesmosomen der dermoepidermalen Junktionszone, identifiziert werden. Der selbstlimitierende Krankheitsverlauf und die hohe Rezidivneigung bei Folgeschwangerschaften deuten auf eine pathogenetische Rolle schwangerschaftsspezifischer Veränderungen endokriner sowie immunologischer Faktoren hin. In der vorliegenden Arbeit wurden therapeutische und immunmodulatorische Effekte einer TNF-Blockade bei Patienten mit aktiver Spondylitis ankylosans untersucht. Hierzu wurde an 10 Patienten während einer 60wöchigen Infliximabtherapie eine Evaluation von Therapiewirksamkeit und -sicherheit, sowie eine durchflusszytometrische Analyse der HLA-Oberflächenexpression auf Lymphozyten vorgenommen. Von 8 Patienten wurde die Therapie ohne ernste Nebenwirkungen gut vertragen und führte zur raschen und dauerhaften Verbesserung der klinischen, laborchemischen und radiologischen Verlaufsparameter. An expressed beta-tubulin gene (TUBB) has previously been localized to chromosome region 6pter-p21 in man. By using a panel of deletion mutant cell lines and radiation-reduced hybrids containing fragments of chromosome 6, the TUBB locus could be mapped to the HLA class I region at 6p21.3. A long range restriction map including TUBB and several HLA class I genes was then generated by rotating field gel electrophoresis. Several new HLA-B (B8, B51, Bw62)- and HLA-C (Cw6, Cw7)-specific genes were isolated either as genomic cosmid or cDNA clones to study the diversity of HLA antigens. The allele specificities were identified by sequence analysis in comparison with published HLAB and -C sequences, by transfection experiments, and Southern and northern blot analysis using oligonucleotide probes.
