DeepSeek’s MHC: Smarter Architecture Over More Compute | 07th Jan 2025
Send us a textHow Parallel Reasoning Channels Are Redefining AI ScalingIn this episode of the Colaberry AI Podcast, we explore a major architectural breakthrough from the Chinese AI firm DeepSeek known as Manifold Constrained Hyperconnections (MHC)—a technique that challenges one of the most persistent limitations in modern AI design. For over a decade, large models have relied on narrow internal information pathways that create bottlenecks for complex reasoning, especially in logic and mathematics. While previous efforts to widen these pathways often led to training instability an...

Artificial Intelligence : Papers & Concepts
DeepSeek mHC
Why do some large AI models suddenly collapse during training—and how can geometry prevent it? In this episode of Artificial Intelligence: Papers and Concepts, we break down DeepSeek AI's Manifold-Constrained Hyperconnections (mHC), a new architectural approach that fixes training instability in large language models. We explore why traditional hyperconnections caused catastrophic signal explosions, and how constraining them to a geometric structure—doubly stochastic matrices on the Birkhoff polytope—restores stability at scale. You'll learn how mHC reduces signal amplification from 3,000× to ~1.6×, enables reliable training of 27B-parameter models, and even improves reasoning performance—all with minimal ov...

DeepSeek: Manifold-Constrained Hyper-Connections (mHC)
This episode explores Manifold-Constrained Hyper-Connections (mHC), a framework designed to solve the training instability and memory overhead issues found in existing Hyper-Connection architectures. We discuss how mHC uses the Sinkhorn-Knopp algorithm to project residual connections onto a doubly stochastic manifold, effectively restoring the "identity mapping" property essential for stable signal propagation. Finally, we examine the infrastructure-level optimizations—such as kernel fusion and selective recomputing—that allow mHC to achieve superior scalability and performance in large-scale foundation models.

Ep. 011 - Digestive Hell
Optimal digestion is crucial for overall health and well-being. It ensures that the body efficiently breaks down food into nutrients, which are then absorbed and utilized for energy, growth, and cellular repair. Proper digestion supports a strong immune system, maintains a healthy gut microbiome, and helps prevent gastrointestinal disorders. Additionally, it plays a significant role in mental health, as the gut is often referred to as the "second brain" due to its impact on mood and cognitive function. Therefore, maintaining optimal digestion is vital for sustaining physical and mental health, enhancing quality of life, and preventing chronic diseases.

Unmasking Leukemia: The Role of MHC:TCR Signaling with Dr. Laffey
This episode delves into the cutting-edge research of Dr. Laffey, who has been investigating the role of Major Histocompatibility Complex:T cell antigen receptor (MHC:TCR) signaling in the development of T cell tumors. While this signaling pathway is crucial for T cell development and immune responses, it can also trigger the formation of T cell tumors under abnormal conditions.Dr. Laffey's research focuses on a low-frequency progenitor cell population known as Early αβ TCR+ Double-Negative (EADN) cells. Present in both mice and humans, these cells can transform into thymic leukemia. His work has shown that EADN cells do...

MHC class I loss in endometrial carcinoma
MHC class I loss by tumor cells decreases tumor neoantigen presentation to the immune system and therefore represents a possible mechanism of immunotherapeutic resistance in cancers that otherwise could be good candidates for checkpoint inhibition (e.g. mismatch repair (MMR)-deficient and PD-L1-positive tumors). In this episode our guest, Dr. Anne Mills discusses her recent Modern Pathology publication on loss of MHC class 1 expression in some endometrial cancers. Study by Friedman et al, MHC class I loss in endometrial carcinoma: a potential resistance mechanism to immune checkpoint inhibition. MHC 29: The Beginning of the End
I uploaded the first episode of MHC on July 16, 2019, and as we reach the one year anniversary of this show, I have also come to the conclusion that we should reach the end as well. Due to the current climate of the world, as well as my own personal uncertainties and feelings, I have decided that this shall be the penultimate episode of MHC. Today I touch on why, as well as on how mental health can affect a person's passions and lead to burnout.

MHC Tetramer
MHC Tetramers have emerged as an important tool for characterization of the specificity and phenotype of T cell immune response, useful in a large variety of disease and vaccine studies. Peptide-MHC multimers display an antigenic peptide in the MHC binding groove, functioning as a surrogate for recognition events that occur as part of the T cell interaction with antigen-presenting cell. The most prevalent form of multimer in use today consists of biotin-labeled pMHC displayed on streptavidin molecules, forming tetravalent complexes; so the common use of the term “tetramers” for this detection method. Since first description, MHC tetramers have become widely used...

Macroautophagy substrates are loaded onto MHC class II of medullary thymic epithelial cells for central tolerance
The immune system is highly organized and multifunctional; it protects the organism against invading pathogens (immune response), while at the same time being tolerant to ‘self’ (immune tolerance). Central tolerance is achieved during T-cell development by negative selection via self-peptide-MHC complexes (pMHC) presented by thymic dendritic cells (tDCs) and medullary thymic epithelial cells (mTECs). However, how self-antigens are processed into peptides that further are shuttled onto major histocompatibility complex (MHC) class II in thymic epithelial cells (TECs) is not fully understood. One candidate pathway involved in this process is macroautophagy (referred to as autophagy hereafter), originally thought to solely be invo...

Analysis of MHC I and II presentation on Equid herpesvirus 1 infected cells Various Alphaherpesviruses are able to interfere with MHC class I presentation by reducing surface expression of these molecules. Equid herpesvirus 1 (EHV-1) infection also results in MHC I downregulation on the respective cells, but other than the involvement of the viral UL49.5-protein, this process is not yet very well understood (Rappocciolo et al., 2003; Ambagala et al., 2004; Koppers-Lalic et al., 2005). As the in vitro non essential EHV-1 proteins UL11p and UL43p were considered as further candidate proteins for playing a role in interfering with MHC I or...

Edição #10: Efeitos na apresentação de antigénios via MHC classe II pelas celulas dendriticas maturas in vivo
Olá, Viva. Décima edição do podcast "Journal Club Imunoterapia tumoral".

Alloreactive cytotoxic T lymphocytes generated in the presence of viral- derived peptides show exquisite peptide and MHC specificity
The nature of alloreactivity to MHC molecules has been enigmatic, primarily because of the observation that allogeneic responses are considerably stronger than syngeneic responses. To better determine the specificity potential of allogeneic responses, we have generated alloreactive CTL specific for exogenous, viral-derived peptide ligands. This approach allowed us to critically evaluate both the peptide- and MHC-specificity of these alloreactive T cells. Exploiting the accessibility of the H-2Ld class I molecule for exogenous peptide ligands, alloreactive CTL were generated that are specific for either murine cytomegalovirus (MCMV) or lymphocytic choriomeningitis virus (LCMV) peptides bound by Ld alloantigens. Peptide specificity was...

EXPRESSION OF A FUNCTIONAL CHIMERIC lg-MHC CLASS II PROTEIN
composed of the a- and ß-chains of the MHC class I1 I-E molecule fused to antibody V regions derived from anti-human CD4 mAb MT310. Expression vectors were constructed containing the functional, rearranged gene segments coding for the V region domains of the antibody H and L chains in place of the first domains of the complete structural genes of the I-E a- and ß-chains, respectively. Celltsr ansfected with both hybrid genes expressed a stable protein product on the cell surface. The chimeric molecule exhibited the idiotype of the antibody MT310 as shown by binding to the anti-idiotypic mAb 20-46. A pr...

A pentapeptide as minimal antigenic determinant for MHC class I-restricted T lymphocytes
Peptides that are antigenic for T lymphocytes are ligands for two receptors, the class I or II glycoproteins that are encoded by genes in the major histocompatibility complex, and the idiotypic / chain T-cell antigen receptor1–9. That a peptide must bind to an MHC molecule to interact with a T-cell antigen receptor is the molecular basis of the MHC restriction of antigen-recognition by T lymphocytes10,11. In such a trimolecular interaction the amino-acid sequence of the peptide must specify the contact with both receptors: agretope residues bind to the MHC receptor and epitope residues bind to the T-cell antigen receptor12,13. From a co...

Involvement of Mhc Loci in immune responses that are not Ir-gene-controlled
Twenty-nine randomly chosen, soluble antigens, many of them highly complex, were used to immunize mice of two strains, C3H and B10.RIII. Lymphnode cells from the immunized mice were restimulated in vitro with the priming antigens and the proliferative response of the cells was determined. Both strains were responders to 28 of 29 antigens. Eight antigens were then used to immunize 11 congenic strains carrying different H-2 haplotypes, and the T-cell proliferative responses of these strains were determined. Again, all the strains responded to seven of the eight antigens. These experiments were then repeated, but this time -antibodies specific for the A...
