Zum Vorkommen von Prion Proteinen in der Milchdrüse und der Milch von Hauswiederkäuern
Die vorliegende Arbeit untersuchte im ersten Teil tierart-vergleichend das Vorkommen von PrPC bei Hauswiederkäuern in Eutergewebe und in den verschiedenen Milchfraktionen. Dies erfolgte mittels EIA und Western-Blot sowie für das Eutergewebe auch mittels Immunhistochemie (IHC). Für die Aufarbeitung von Rahm wurde dabei ein neues Verfahren etabliert, durch das die Proteinfraktion des Rahms als Pellet gewonnen werden konnte. Viele neurodegenerative Erkrankungen, wie die transmissiblen spongiformen Enzephalopathien (TSE), die Alzheimer- und die Huntington-Krankheit, sind durch charakteristische Ablagerungen im Gehirn, sogenannte Amyloide, gekennzeichnet. Amyloide sind oftmals fibrilläre Aggregate von normalerweise löslichen Proteinen, deren dreidimensionale Strukturen sich bei der Aggregation verändern. Bedauerlicherweise waren hochauflösende Methoden biophysikalischer Strukturaufklärung bislang auf Amyloide nicht anwendbar. Zum Nachweis von pathogenen Prionen mittels "Kapillarelektrophorese-laserinduzierter Fluoreszenz (CE-LIF)"
Zum Nachweis von PrPc und PrPsc wurde eine Kapillarelektrophorese-Immunofluoreszenz-Methode (CE-LIF) etabliert und mit konventionellen Verfahren (ELISA, Westernblot, Dot-EIA) verglichen. Zur CE-LIF wurde der monoklonale Antikörper F89 und ein Fluoreszenz-markiertes Peptid (FITC-P3) eingesetzt. Die Nachweisgrenze für rekombinantes bovines PrPc liegt bei 1 pg/Test. Molekulare Mechanismen der Prionpropagation
Ein zentrales Ereignis in der Pathogenese der Prionkrankheiten ist die strukturelle Konversion des zellulären Prionproteins (PrPC) in eine infektiöse Isoform (PrPSc). Im Rahmen der vorliegenden Arbeit wurde die von der Prion-Hypothese angenommene Eigenschaft von PrPSc zur autokatalytischen Vermehrung durch serielle PMCA-Reaktionen experimentell belegt und die Vermehrung von Infektiösität in vitro dargestellt. Die Fragmentierung der PrP-Aggregate in kleinere Aggregationskeime hat sich dabei für eine effiziente autokatalytische Prion-Replikation als essentiell erwiesen. Mit dem Testverfahren wurde auch Milch von Rindern, Ziegen und Schafen auf das Vorkommen von PrPC und dessen Verteilung auf die einzelnen Milchfraktionen untersucht.
