Are Cell Phone Carrier Trade-in Deals a Trap?
Every fall, wireless carriers flood the market with irresistible offers: "$800 off a new phone with a trade-in!" It feels like a massive discount, but we’re here to expose the truth: You are caught in a brilliantly rigged financial game. This episode cuts through the carrier marketing to reveal the actual total cost of ownership (TCO) for your smartphone. We’ll show you why frequently upgrading your device—even with a high trade-in credit—is the most expensive mistake you can make, and how the lowest annualized cost is achieved by simply keeping your phone for 3 to 4 years.

Hematuria in the Hospitalized Patient: Master the Evidence-Based Approach to Risk, Workup, and The Anticoagulation Trap
In this episode of Hospital Medicine Unplugged, we sprint through hematuria in the hospital—classify fast, stabilize what’s dangerous, risk-stratify smartly, and image with purpose. We open with the do-firsts: confirm gross vs. microscopic (≥3 RBC/HPF) on a proper urinalysis; repeat if contamination or a transient cause is likely. Don’t blame anticoagulation—it can unmask disease, not explain it away. Take a tight history (infection, stones, trauma/instrumentation, menstruation, exercise, meds, malignancy risks) and exam (vitals, bladder distention, CVA tenderness, external sources). Red flags that change the room: clot retention/obstruction, hemodynamic instability, sepsis, AK...

Logan Soule on Blood Storage: Even Red Blood Cells Get a Sugar Crash
Blood transfusions can be life-saving procedures for patients in need. However, they can also impart much more devastating complications, leading to poorer outcomes than prior to transfusion, such as infection, inflammation, and even death. This is in part due to the red blood cell “storage lesion”, which refers to the irreversible metabolic and physiological damages that occur to red blood cells (RBCs) during their storage period, such as membrane damage, protein/lipid oxidation, glycation, cell lysis, and many other detrimental changes vital to proper RBC function. Logan Soule is a 5th year PhD candidate in the Department of Biomedical Engi...

But over recent years, immunologist Nilam Mangalmurti, University of Pennsylvania, has been finding several clues to challenge that dogma – including molecules on the surface of red blood cells known from other parts of the immune system. Blood Groups
Blood can be grouped by using two groupings ABO grouping, Rh grouping. ABO Grouping Antigens are present on the surface of the RBCs .Therefore, ABO grouping depends on the presence of type of antigen on the RBCs, and namely antigens are only A, B. So blood groups are A, B, AB, and O. A blood group have only A antigens on the surface of the RBC and B blood group have only B antigens on the surface of the RBCs . Since AB blood group have both A and B antigens on the surface...

What to do when serum magnesium is high but RBC magnesium is low? | Masterjohn Q&A Files #79
Question: What to do when serum magnesium is high but RBC magnesium is low? The magnesium in the blood and the hair is high. When you say blood, I'm assuming this is serum or plasma because the RBC magnesium is low. I'm hoping that's not whole blood magnesium in which case it would be hard to separate from the RBC magnesium. But I mean even for whole blood, if the RBC magnesium is low and the blood magnesium is high, then the magnesium that's in the blood that's high is in the serum or plasma, not in...

Structural und functional characterization of Rubisco assembly chaperones
In the present study, the structure and mechanism of two assembly chaperones of Rubisco, Raf1 and RbcX, were investigated. The role of Raf1 in Rubisco assembly was elucidated by analyzing cyanobacterial and plant Raf1 with a vast array of biochemical and biophysical techniques. Raf1 is a dimeric protein. The subunits have a two-domain structure. The crystal structures of two separate domains of Arabidopsis thaliana (At) Raf1 were solved at resolutions of 1.95 Å and 2.6–2.8 Å, respectively. The oligomeric state of Raf1 proteins was investigated by size exclusion chromatography connected to multi angle light scattering (SEC-MALS) and native mass spectrometry (MS). Both cyanobacterial and plan...

Microcirculatory assessment of red blood cell transfusion in children with severe anemia
Background: Pediatric hematology patients frequently receive red blood all transfusions for severe anemia. Our goal was to assess the effect of blood transfusion on the microcirculation and thus provide information on the quality of tissue perfusion. Methods and patients: The sublingual microcirculation was visualized with Sidestream-Darkfield Imaging in 20 anemic (Hb: 7.2 g/dL, 95% CI 6.6-7.9) children receiving red blood cell transfusions and in age matched healthy non-anemic controls. Functional vessel density (FVD) was determined with a semiautomatic program. Results: Immediately after transfusion FVD increased (13.4 versus 15 mm/mm2) and RBC velocity (696 (598-792) versus 628 (549-707) μm/s) decreased but FVD was always significantly l...

Significance of Azathioprine Metabolites for the Maintenance of Remission in Autoimmune Hepatitis
Drs. Stephen Harrison and Harpreet Dhaliwal Azathioprine (AZA) is used to maintain remission in autoimmune hepatitis (AIH), but up to 18% of patients are unresponsive. AZA is a prodrug, and the formation of active thioguanine nucleotide (TGN) metabolites varies widely. We aimed to assess the relationship between AZA metabolite concentrations (i.e., TGNs and methylmercaptopurine nucleotides [MeMPNs]), thiopurine methyltransferase (TPMT) activity, therapeutic response, and toxicity in adult patients with AIH prescribed a stable dose of AZA for the maintenance of remission. Red blood cell (RBC) TGNs and MeMPNs were measured in serial blood samples over a 2-year period. The average TGNs...

Computational identification of phospho-tyrosine sub-networks related to acanthocyte generation in neuroacanthocytosis
Acanthocytes, abnormal thorny red blood cells (RBC), are one of the biological hallmarks of neuroacanthocytosis syndromes (NA), a group of rare hereditary neurodegenerative disorders. Since RBCs are easily accessible, the study of acanthocytes in NA may provide insights into potential mechanisms of neurodegeneration. Previous studies have shown that changes in RBC membrane protein phosphorylation state affect RBC membrane mechanical stability and morphology. Here, we coupled tyrosine-phosphoproteomic analysis to topological network analysis. We aimed to predict signaling sub-networks possibly involved in the generation of acanthocytes in patients affected by the two core NA disorders, namely McLeod syndrome (MLS, XK-related, Xk protein...

Rubisco folding and oligomeric assembly: Detailed analysis of an assembly intermediate
To become biologically active, a protein must fold into a distinct three-dimensional structure. Many non-native proteins require molecular chaperones to support folding and assembly. These molecular chaperones are important for de novo protein folding as well as refolding of denatured proteins under stress conditions. A certain subset of chaperones, the chaperonins, are required for the folding of the enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco); furthermore, correct folding of Rubisco is also aided by the Hsp70 chaperone system. Rubisco catalyzes the initial step of CO2 assimilation in the Calvin-Benson-Bassham (CBB) cycle. Unfortunately, this enzyme is extremely inefficient, not only does it...

Hemotrophic Mycoplasmas Induce Programmed Cell Death in Red Blood Cells
Hemotrophic mycoplasmas (HM) are uncultivable bacteria found on and in the red blood cells (RBCs). The main clinical sign of HM infections is the hemolytic anemia. However, anemia-inducing pathogenesis has not been totally clarified. In this work we used the splenectomized pig as animal model and Mycoplasma suis as a representative for hemotrophic mycoplasmas to study anemia pathogenesis. Eryptosis, i.e. programmed cell death of RBCs, is characterized by cell shrinkage, microvesiculation and phosphatidylserine (PS) exposure on the outer membrane. The eryptosis occurrence and its influence on anemia pathogenesis was observed over the time-course of M. suis infections in pigs...

Folding and assembly of RuBisCO
To become biologically active, proteins have to acquire their correct three-dimensional structure by folding, which is frequently followed by assembly into oligomeric complexes. Although all structure relevant information is contained in the amino acid sequence of a polypeptide, numerous proteins require the assistance of molecular chaperones which prevent the aggregation and promote the efficient folding and/or assembly of newly-synthesized proteins. The enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO), which catalyzes carbon fixation in the Calvin-Benson-Bassham cycle, requires chaperones in order to acquire its active structure. In plants and cyanobacteria, RuBisCO (type I) is a complex of approximately 550 kDa composed of...

Platelet kinetics in the pulmonary microcirculation in vivo assessed by intravital microscopy
Growing evidence supports the substantial pathophysiological impact of platelets on the development of acute lung injury. Methods for studying these cellular mechanisms in vivo are not present yet. The aim of this study was to develop a model enabling the quantitative analysis of platelet kinetics and platelet-endothelium interaction within consecutive segments of the pulmonary microcirculation in vivo. New Zealand White rabbits were anesthetized and ventilated. Autologous platelets were separated from blood and labeled ex vivo with rhodamine 6G. After implantation of a thoracic window, microhemodynamics and kinetics of platelets were investigated by intravital microscopy. Velocities of red blood cells (RBCs...
