跟随肿瘤界,听点肿瘤学。大家好,这里是肿瘤界100s,我是主播李静。书接上回,我们继续mTOR的故事。直到90年代,人们首先在酵母菌中发现了雷帕霉素的靶点TOR。随后在哺乳动物中发现了与雷帕霉素的结合蛋白,由于其与酵母TOR基因有同源性,因此在TOR的前面加上了定语哺乳动物,最终定名为mTOR,m是英文哺乳动物mammal的首字母,即哺乳动物。那为什么mTOR还有个FRAP1的怪名呢?因为当时还有一款和雷帕霉素作用机制差不多的免疫抑制剂,叫做FK506,有人从大肠杆菌中筛选出了一款与FK506结合的蛋白质,起名FK506结合蛋白12(FK506-binding protein 12),进一步研究发现该蛋白质也能和雷帕霉素结合,就叫它FK506结合蛋白12-雷帕霉素相关蛋白1(FK506-binding protein 12-rapamycin-associated protein 1 ),名字是不是特别长,而且很拗口,它的缩写就是FRAP1,也就是mTOR。可见TOR基因确实是一个古老的基因,在酵母、细菌中都有存在,甚至在植物中也有重要的生长调节作用,到了哺乳动物中也并没有多大的改变。其实,FRAP1发现在前,mTOR发现在后,为什么mTOR称为正式称呼了呢?这还得感谢德语。在德语中,TOR的意思是“门”或“门户”,更能突出TOR的代谢调节功能。2009年,HUGO基因命名委员会正式将FRAP1基因名称更改为mTOR。现在已经明确,mTOR是PI3K相关激酶家族的一员,是细胞生长和增殖的重要调节因子。mTOR信号的过度激活会助长包括乳腺癌、前列腺癌、肺癌等多种肿瘤的发生和发展,抑制mTOR也在临床被证实有助于改善部分肿瘤患者的预后。基于mTOR调节多种细胞信号传导通路和代谢的作用,近年来学术界决定将mTOR的起始字母m的含义改为mechanistic,代表它具有调节细胞机制的功能,而不仅仅是雷帕霉素的一个靶点。mTOR的故事就到这里了,我是李静,感谢您的收听,我们下期见。

跟随肿瘤界,听点肿瘤学。大家好,这里是肿瘤界100s,我是主播李静。前两天我们在CAPItello研究那一期,提到了PI3K-AKT-mTOR通路,留了一个关于mTOR的小尾巴,那么今天我们一起mTOR通路的神秘面纱。mTOR抑制剂常作为HR+/HER2-的乳腺癌内分泌治疗的联合用药,也可用于晚期肾癌和部分神经内分泌肿瘤。单看mTOR(mammalian Target Of Rapamycin)这个名字有点让人一知半解,直译过来就是哺乳动物雷帕霉素靶蛋白,名字里还有一种抗真菌药物的名字,这是怎么回事?而且mTOR在有的文献里还被称为FRAP1,而且mTOR开头的m也有不同的含义,那么本期节目我们就来一一答疑解惑。故事还要从1960年代讲起,当时有人去复活节岛探险,这个与世隔绝的岛屿具备孕育天然抗生素的土壤。1972年,一位名叫Sehgal的学者从岛上土壤样本中的链霉菌内发现了一种小分子,将其纯化后证实这一分子具有抗真菌活性。Sehgal将这种抗真菌分子命名为雷帕霉素。因为复活节岛在本地语中叫做Rapa Nui,加上链霉菌的拉丁名Streptomyces,取后缀加上rapa,拼起来就是雷帕霉素。这也是我们的主角mTOR中雷帕霉素的由来。雷帕霉素除了抗真菌作用外,还被发现有免疫抑制和抗细胞增殖的作用。雷帕霉素在1980年代受到制药行业的极大兴趣,FDA曾批准雷帕霉素作为肾移植后的免疫抑制剂,但当时并不清楚雷帕霉素的作用机制。时间所限,本期100s就到这里了,我是李静,感谢您的收听,我们下期继续谈谈关于mTOR的故事。
